Exposure to fine particles can cause various diseases, and an easily accessible method to monitor the particles can help raise public awareness and reduce harmful exposures. Here we report a method to estimate PM air pollution based on analysis of a large number of outdoor images available for Beijing, Shanghai (China) and Phoenix (US). Six image features were extracted from the images, which were used, together with other relevant data, such as the position of the sun, date, time, geographic information and weather conditions, to predict PM2.5 index. The results demonstrate that the image analysis method provides good prediction of PM2.5 indexes, and different features have different significance levels in the prediction. 相似文献

A total of 15 polymorphic tetranucleotide microsatellite loci in the red panda, Ailurus fulgens, were characterized in this study. Based on evaluations of 33 red pandas, the number of observed alleles for each locus ranged from seven to 17 and the expected and observed heterozygosities were 0.412–0.897 and 0.121–0.909, respectively. The mean polymorphic information content was 0.721. These markers would greatly strengthen the utilization of microsatellite tools in genetic variation studies in red panda populations. 相似文献

The native serine protease proteinase K binds two calcium cations. It has been reported that Ca2+ removal decreased the enzyme’s thermal stability and to some extent the substrate affinity, but has discrepant effects on
catalytic activity of the enzyme. Molecular dynamics simulations were performed on the Ca2+-bound and Ca2+-free proteases to investigate the mechanism by which the calciums affect the structural stability, molecular motions, and
catalytic activity of proteinase K. Very similar structural properties were observed between these two forms of proteinase
K during simulations; and several long-lived hydrogen bonds and salt bridges common to both forms of proteinase K were found
to be crucial in maintaining the local conformations around these two Ca2+ sites. Although Ca2+ removal enhanced the overall flexibility of proteinase K, the flexibility in a limited number of segments surrounding the
substrate-binding pockets decreased. The largest differences in the equilibrium structures of the two simulations indicate
that, upon the removal of Ca2+, the large concerted motion originating from the Ca1 site can transmit to the substrate-binding regions but not to the catalytic
triad residues. In conjunction with the large overlap of the essential subspaces between the two simulations, these results
not only provide insight into the dynamics of the underlying molecular mechanism responsible for the unchanged enzymatic activity
as well as the decreased thermal stability and substrate affinity of proteinase K upon Ca2+ removal, but also complement the experimentally determined structural and biochemical data. 相似文献

Opioids, like other drugs, are thought to initiate their effects by association with their specific receptors. However, very little is known about the opioid receptor as a molecular entity. The binding components have been solubilized in detergent and purified by different approaches, but the molecular size of soluble opioid receptor complexes reported by different groups varied from 23,000 to 750,000. In this study, the technique of radiation inactivation by gamma rays was used to investigate the apparent size of the opioid receptor in rat brain membranes under different conditions. The molecular sizes of opioid receptor complexes were estimated as 313,000 +/- 13,500 in the presence of [D-Ala2, D-Leu5] enkephalin, NaCl and Gpp (NH)p; as 165,000 +/- 8,500 in the presence of NaCl only, or of both NaCl and Gpp (NH)p; as 217,000 +/- 6,600 in the presence of Gpp (NH)p only; and as 286,000 +/- 60,900 in the presence of MgCl2 only. A simple model has been proposed to explain these different apparent target sizes of opioid receptors obtained under different conditions. 相似文献

人工微生物混菌系统的生物工程应用价值日益受到重视,使得对于混菌系统中成员菌间的相互作用机制研究也成为近年来的一个热点。其研究结果一方面可以为现有人工混菌系统的进一步优化提供理论依据,另一方面也为全新混菌系统的人工构建提供新的思路和策略,进而促进人工微生物混菌系统未来规模化应用。基因组学、转录组学、蛋白质组学和代谢组学等研究方法能够高通量分析各种生物分子、提供大量的数据与信息,多组学分析可以获得混菌系统中细胞的“全景”,在揭示人工微生物混菌系统中各个成员间的相互作用的研究中有着特殊的意义。文中综述了近年来多种组学技术在人工微生物混菌系统机制解析中的应用及研究进展,从代谢网络、能量代谢、信号转导、膜转运、胁迫响应、混菌系统的稳定性以及结构合理性等方面探讨混菌系统机制解析的最新进展,以期为利用合成生物学、基因组编辑等新兴生物技术改造微生物混菌系统实现其工程化应用提供理论依据。 相似文献

N-糖蛋白去糖基化酶(PNGase)是一种广泛存在于真菌、植物、哺乳动物中的去糖基化酶,可以水解N-糖蛋白或 N-糖肽上天冬酰胺与寡糖链连接的化学键,并释放出完整的N-寡糖。PNGase在生物体内参与蛋白质降解、器官发育、个体生长等过程。人PNGase基因功能缺陷会导致先天性去糖基化障碍,小鼠PNGase缺陷会导致胚胎致死性,线虫PNGase缺陷使其寿命下降。本文对PNGase在不同物种的分布、蛋白质结构、酶学功能及生物学功能进行阐述,为PNGase的生理病理功能及致病机制的基础研究提供思路,为PNGase作为糖生物学工具酶或药物开发的创新应用研究奠定基础。 相似文献
